The antibody has a proven strong staining using a 1/2000-1/3000 dilution with biotin-streptavidin/HRP technique in human stomach/antrum.

The specificity of the antiserum was evaluated by soluble pre-adsorption with the peptides in question at a final concentration of 10-6 M. Gastrin-17 immunolabeling was completely abolished by pre-adsorption with Gastrin 1 and big Gastrin and partially abolished by CCK-8. Pre-adsorption with the following peptides resulted in no loss of immunostaining: motilin, secretin, PHI-27, bombesin, VIP and GIP.

Photo Description: IHC image of parietal cells in the human stomach staining for gastrin (above, low magnification image; below high magnification image). The tissue was formalin fixed and embedded in paraffin for microtome sectioning. Sections at RT in 10% goat serum in PBS, followed by 3% hydrogen peroxide before standard IHC procedure. Primary antibody was incubated at 1:3000 for 48 hours, goat anti-rabbit secondary was subsequently added for 1 hour after washing with PBS. Light microscopy staining was achieved with standard biotin-streptavidin/HRP procedure and DAB chromogen and hemotoxylin counterstain in the low magnification image.
